چکیده

Background The clinical and economic impact of severe drug hypersensitivity is huge for both healthcare and pharmaceutical industry; however, no method for predicting culprit candidate t drugs under clinical development exists currently. As genetic variants in human-leukocyte antigens (HLA) have been linked to inter-individual differences in the risk of drug hypersensitivity, we ascertained whether HLA library can serve as a drug screening tool for their potential to cause severe drug hypersensitivity.

Epidemiology of Severe Cutaneous Adverse Drug Reaction and Its HLA Association among Pediatrics

Severe cutaneous adverse drug reaction (SCAR) is considered to be a multifactorial drug side effect. This study was designed to investigate the epidemiology and human leukocyte antigen (HLA)-A and -B gene polymorphisms in pediatric patients with SCAR admitted in tertiary referral center, southwestern of Iran from 2013 to 2017. Demographic data, past allergy and autoimmune history, clinical pres...

A prospective study of HLA*B-5801 genotyping in preventing allopurinol- induced severe cutaneous adverse reactions

Background Allopurinol, a commonly prescribed medication for gout and hyperuricemia, is a frequent cause of severe cutaneous adverse reactions (SCAR), which include the hypersensitivity syndrome, Stevens-Johnson syndrome, and toxic epidermal necrolysis. Allopurinol-induced SCAR is strongly associated with the HLA-B*58:01 allele. We sought to prevent allopurinol-induced SCAR by using HLA-B*58:01...

HLA and pharmacogenetics of drug hypersensitivity.

Immunologically mediated drug reactions have been traditionally classified as unpredictable based on the fact that they cannot be predicted strictly on the pharmacological action of the drug. Such adverse drug reactions are associated with considerable morbidity and include severe cutaneous adverse reactions such as Stevens-Johnson syndrome/toxic epidermal necrolysis and the drug hypersensitivi...

HLA Associations and Clinical Implications in T-Cell Mediated Drug Hypersensitivity Reactions: An Updated Review

T-cell mediated drug hypersensitivity reactions may range from mild rash to severe fatal reactions. Among them, drug reaction with eosinophilia and systemic symptoms (DRESS) or drug-induced hypersensitivity syndrome (DIHS), Stevens-Johnson syndrome/ toxic epidermal necrolysis (SJS/TEN), are some of the most life-threatening severe cutaneous adverse reactions (SCARs). Recent advances in pharmaco...
